Active compound

Hyperforin

phloroglucinol derivative

The main mood-active constituent of St John's wort — and the reason it interacts with so many drugs.

Mechanism

Inhibits reuptake of serotonin, norepinephrine, dopamine, GABA, and glutamate; also potently induces the drug-metabolising enzyme CYP3A4 via the pregnane X receptor, which is what depletes many co-taken medicines.

monoamine reuptake pregnane X receptor CYP3A4
